I am working on a Vibrverb 'like' build… very close to the Encore. Is it necessary and advised to isolate all four RCA jacks that connect to the Tremolo connection to the floor pedal, Reverb jacks to and from the pan, and the jack for the reverb button on the floor pedal?

Millions of Fenders & other amps around, with no ground isolation of the RCA jacks to tank & pedal. They all work just fine. Save yourself the trouble.

My best friend and I built matching Deluxe Reverbs and I isolated all my jacks except for the speakers. He followed the original ground scheme and had a small amount of audible hum where mine did not.
Even though it wasn't real bad, it bugged the hell out of him until he copied mine.

Joe, yes on that your right and that is why on two channel Fenders like that when you turn up the controls some on the normal channel that hum level your friend had will be reduced, as in phase canceled due to the two channels BEING out of phase with each other.
