I do. Just in case but it pretty obvious when the nut gets loose and the sound cuts in and out. Same with pots. If your going to build, you might as well go the extra mile?

I use the black plastic jacks, which are not grounded in themselves. I was wondering if it's necessery to ground one side to chassis/grounding point, or doesn't it have to be?

In another amp I made, I haven't grounded anything on the secondary sie of the OT, and it works fine...

I also use the black plastic jacks, and I simlpy bring the ground to of my other ground points. Inputs go to preamp gound point, outputs go to power stage ground point, etc.

Yes it should be
If you have negative feedback it must be grounded.
IF not you still should...
If there is an OT fault your speakers could wind up with your B+ on them.
An ungrounded secondary will be capacitively coupled to the primary, can have a high AC swing which can make it's way back to the input and cause other crap like squeals. The core should also be grounded to chassis as well as your choke and power transformer through their mounting bolts.
